Indications

Vitamin B complex is recommended for preventative or therapeutic nutritional supplementation in physiologically demanding settings. Glossitis, stomatitis, cheilosis, and beriberi are some of the conditions that cause B-vitamin deficiency or impaired absorption or bioavailability. Pharmacology

The vitamin B complex contains the most important elements of the vitamin B group in pure form and in therapeutically adjusted amounts. Members of the vitamin B group are components of enzyme systems that regulate various stages of carbohydrate, lipid, and protein metabolism, with each component serving a distinct biological function. B vitamin deficiency symptoms include glossitis, stomatitis, cheilosis, polyneuritis, beriberi, pellagra, and corneal vascularization.

Dosage & Administration

• The usual starting dose for a film-coated tablet is 40 to 80 mg daily. When necessary, the dose can be increased to 320 mg daily in divided doses. The medication should be taken before eating. Gliclazide is not used in children because it is contraindicated in juvenile-onset diabetes.
• The usual daily dosage for Gliclazide 30 mg is 1 to 4 tablets per day as a single dose, and for Gliclazide 60 mg is 1 to 2 tablets per day as a single dose. It is suggested that the medication be taken in the morning. It is preferable to take this medication before breakfast to avoid or minimize the onset of digestive disorders.

Interaction

Gliclazide's hypoglycemic effects may be exacerbated by aspirin, NSAIDs, phenylbutazone, clofibrate, sulfonamide, cimetidine, imidazole antifungal medications, and monoamine oxidase inhibitors. Gliclazide's hypoglycemic effect may be reduced by Rifampin, Barbiturates, Phenytoin, Thiazide diuretics, Diazoxide, Glucocorticoids, Estrogens, Sympathomimetic medications, and Alcohol.

Contraindications

The following conditions should not be treated with gliclazide: Diabetes of juvenile-onset (type 1) Diabetes in conjunction with Ketosis Diabetic patients undergoing surgery, recovering from a serious accident, or suffering from an infection Patients who are allergic to the sulfonylurea class of medications Breast-feeding mothers and pregnant women Inadequate liver function.

Side Effects

There may be hypoglycaemia in concurrent conditions such as hepatic and renal disease, malnutrition, alcohol intoxication. Sometimes headache, gastrointestinal upsets, nausea, dizziness, skin reactions including rash, pruritus, erythema may occur.

Pregnancy & Lactation

Gliclazide is contraindicated in pregnant women. It should not be used in breast feeding mother.

Precautions & Warnings

Gliclazide should be used carefully in patients with hepatic impairment. If there is definite hepatic disease then gliclazide should not be used. Gliclazide can be used safely in patients with renal insufficiency as it is extensively metabolized. Gliclazide has less sudden hypoglycaemic episode than other sulfonylurea group of drugs
